I. Background | |
B 细胞成熟抗原 (BCMA) 是一种跨膜糖蛋白,属于肿瘤坏死因子受体(TNFR)超家族,表达于正常、恶性浆细胞和成熟 B 细胞表面,其他组织中表达量极少。BCMA 与另外两个功能相关的TNFR 超家族成员—B 细胞活化因子受体(BAFF-R)和 TACI 一起,协同调节 B 细胞增殖、成熟和存活,以及分化为浆细胞 (PC)。BCMA具有一种可溶形式 sBCMA,其通过 γ-分泌酶活性直接从膜 BCMA 脱落。sBCMA 保留了胞外结构域和部分跨 膜区。sBCMA 是 B 细胞参与人类自身免疫性疾病(如系统性红斑狼疮、类风湿性关节炎 和多发性硬化症)的潜在生物标志物。 BCMA 有两种激动剂配体:增殖诱导配体 (APRIL) 和 BAFF;APRIL对 BCMA 的结合亲和力比 BAFF高得多,并且它还能与 TACI 结合,而 BAFF对BAFF-R的选择性更高。 配体与 BCMA结合后,多发性骨髓瘤细胞中多个生长和存活信号级联被激活,最常见的是 活化 B 细胞的核因子 κ 轻链增强子 (NF-κB),但也包括 RAS/MAPK 和 PI3K-Akt 信号通路。这些通路通过调节细胞周期检查点来刺激增殖,通过上调抗凋亡蛋白(如 Mcl-1、 BCL-2、BCL-XL)来提高存活率,以及产生细胞粘附分子(如 ICAM-I)、血管生成因子(如 VEGF、IL-8)和免疫抑制分子(如 IL-10、PD-L1、TGF-β)。 |
