I. Background | |
癌症相关炎症的特征是存在特定的炎症细胞和炎症介质,包括细胞因子和趋化因子。 最近的证据表明,信号转导和转录激活因子(STAT)家族蛋白,特别是 STAT3,在恶性转 化开始和癌症进展过程中,在选择性诱导和维持癌前炎症微环境中起着至关重要的作用。 信号转导和转录激活因子 3 (STAT3)是一种转录激活因子,它将细胞外受体的信号直 接传递到细胞核,介导一些基因的表达,在细胞凋亡等细胞过程中发挥重要作用。在细胞 因子和生长因子的作用下,STAT3 被受体相关 Janus 激酶(JAK) 磷酸化,形成同二聚体或异 二聚体,并转位到细胞核中,作为转录激活剂发挥作用。STAT3 信号传导是恶性肿瘤的主 要内在途径,因为它在恶性细胞中频繁激活,并且在调节肿瘤微环境中对癌症发炎至关重 要的许多基因中起关键作用。 |

STAT3-Luc HEK293 报告基因细胞,是由 STAT3 调控并表达 Luc 荧光素 酶报告基因的HEK293细胞。IL6细胞因子与受体复合物结合,启动JAK/STAT3信号通路, 原理见下图所示。
